Poruchy lipidové biosyntézy v CNS a metabolizmu plazmatických lipidů významným způsobem zasahují již do embryonálního vývoje jedince, jsou původcem celé řady vrozených malformací, neurometabolických a neurodegenerativních onemocnění, hrají roli v procesu aterogeneze. Závažnost klinických projevů je důsledkem podílu genetické výbavy a složky enviromentální. Dynamický rozvoj poznání, zejména v posledním desetiletí přinesl celou řadu poznatků, které změnily pohled na široké spektrum neurologických chorob. V mnoha případech přineslo toto poznání jisté pozitivní terapeutické konsekvence. Práce uvádí přehled vybraných zástupců chorob souvisejících s poruchou cholesterolové homeostázy z oblasti metabolických malformačních syndromů, neurometabolických a neurodegenerativních onemocnění, zmíněn je také podíl dyslipoproteinémií u cerebrovaskulárních onemocnění.
