Dávkování valproátu a jeho vztah k teratogenicitě

Ačkoli se obvyklá účinná dávka valproátu pohybuje v širokém rozmezí mezi 500–2500 mg/den, v klinické praxi je mnoho pacientů kompenzováno na dávkách do 1000 mg/den, zejména těch trpících generalizovanými záchvaty. Z provedeného dotazníkového průzkumu vyplývá, že 1000 mg/den je i průměrná dávka valproátu používaná v ČR při léčbě pacientů se záchvaty parciálními a/nebo sekundárně generalizovanými. Data dostupná v současné době z těhotenských registrů naznačují, že riziko teratogenního vlivu valproátu je na dávce závislé a významně se zvyšuje při použití denních dávek nad 1000–1100 mg/den.
